NEW UPLC and HPLC Columns for Separating Carbohydrates

Waters NEW ACQUITY UPLC BEH Amide columns and XBridge Amide HPLC columns retain polar analytes such as carbohydrates (saccharides) and metabolites that are too polar to retain using conventional reversed-phase LC. Based on Waters novel ethylene bridged hybrid [BEH] particle technology, the 1.7 µm BEH Amide UPLC columns and 3.5 µm XBridge Amide HPLC columns allow routine use of a wide range of mobile phase pH [2 – 11] to promote retention of a wide variety of polar analytes spanning a wide range in polarity, structural moiety and pKa. In addition, since ACQUITY UPLC BEH Amide columns and XBridge Amide columns are based upon the same particle and bonding technologies, they provide the same selectivity, thus allowing scientists to seamlessly transfer their separations between HPLC and UPLC technology platforms.
In addition to enhanced retention of polar compounds, ACQUITY UPLC BEH Amide columns and XBridge Amide HPLC columns provide increased mass spectrometry response, direct compatibility with sample preparation eluates [PPT, LLE and SPE] as well as orthogonal selectivity compared to reversed-phase materials, making HILIC an attractive alternative separation technique.
Classical problems encountered when analyzing carbohydrates include salt interferences, anomer mutarotation, Schiff Base formation, loss of reducing sugars at elevated temperatures, shortened column lifetime, and long analysis times. When using the extremely stable and efficient ACQUITY UPLC BEH Amide columns or XBridge Amide HPLC columns for carbohydrate analysis, none of the problems are encountered.
