New HPLC columns to accelerate advanced therapy research

Thermo Fisher Scientific has announced the release of the SurePac Bio 550 SEC MDi and DNAPac RP semi-preparative columns. These new columns are designed to support the modern laboratory's analytical, efficiency, and scalability needs, particularly in the development of advanced therapies.

The Thermo Scientific SurePac Bio 550 SEC MDi columns feature 3 µm monodisperse particles and inert (MDi) column hardware, offering superior retention of adeno-associated virus (AAV) monomers and higher resolution of aggregates in cell and gene therapy applications. These columns enable fast run times and achieve optimal separation efficiency, providing reliable results that can accelerate the delivery of novel therapeutics to the market.

The Thermo Scientific DNAPac RP semi-preparative columns are designed to aid laboratories in the expanding field of oligonucleotide therapies and precision medicine. Their unique chemistry delivers unmatched performance across a wide range of pH, temperature, and mobile phase compositions, ensuring precise separations and sample purification.

The SurePac Bio 550 SEC MDi columns are ideal for gene therapy and biopharmaceutical analytical laboratories that require size exclusion chromatography (SEC) for determining aggregates in AAV analysis. The DNAPac RP semi-preparative columns are suited for laboratories working with synthetic oligonucleotides, siRNA, ASO, and mRNA, enabling lab-scale purification of both small and large DNA and RNA oligonucleotides.

The SurePac Bio 550 SEC MDi columns offer enhanced efficiency and resolution due to high-resolution separation of AAV and aggregates, thanks to monodisperse particles that improve the homogeneity of packing and reduce peak broadening effects. MDi technology ensures consistent particle size distribution for column-to-column and batch-to-batch reproducibility, achieving consistent results and robust, easy-to-validate methods. The column’s 3 µm particles allow for the use of shorter columns to achieve the desired resolution compared to traditional 5 µm technologies, unlocking significant throughput improvements with faster run times and the ability to process more samples.

The DNAPac RP semi-preparative columns achieve superior reversed-phase resolution and efficient separation in lab-scale purifications. The particles’ wide operational pH range (0-14) and high-temperature range (up to 110 °C) make it ideal for supporting method development. The column chemistry is based on a super-macroporous structure with 4 µm DVB particles and a pore size range of up to 2000 Å, offering excellent separation of both short and long single- and double-stranded nucleic acids, including RNA modalities such as intact mRNA.
