From Bottlenecks to Breakthroughs with High-Throughput DLS

One of the primary technologies for assessing the aggregation, stability, and viscosity of biotherapeutics, ranging from proteins and peptides to attenuated viruses, is dynamic light scattering (DLS). DLS measures particle size and size distributions from less than a nanometer and up to several microns under native conditions without sample modification. The measurement takes place in seconds, and the sample is fully recoverable because the sample remains unperturbed.

 The biotherapeutic pipeline and the molecules being developed are only becoming increasingly numerous and more complicated, necessitating more tests and instrumentation for their interrogation. This situation could create a bottleneck in the development process without automation.

Early in the discovery process, the DynaPro Plate Reader can be deployed as a pre-screening tool before binding assays. Aggregates often bind non-specifically to a target and are detected as false positives. By identifying the presence of aggregates beforehand, DLS serves as a guardian against erroneous results. 

Purification or buffer exchange steps can trigger self-association because of rapid pH or buffer change. Plate-based DLS proves instrumental in process development by quickly sampling these different conditions or monitoring changes over time. 

The DynaPro Plate Reader provides invaluable insights on various stability metrics during formulation development. Its excellent thermal control includes information such as the onset of aggregation and accelerated storage assessment.
