From Bottlenecks to Breakthroughs with High-Throughput DLS
Jun 04 2024
One of the primary technologies for assessing the aggregation, stability, and viscosity of biotherapeutics, ranging from proteins and peptides to attenuated viruses, is dynamic light scattering (DLS). DLS measures particle size and size distributions from less than a nanometer and up to several microns under native conditions without sample modification. The measurement takes place in seconds, and the sample is fully recoverable because the sample remains unperturbed.
The biotherapeutic pipeline and the molecules being developed are only becoming increasingly numerous and more complicated, necessitating more tests and instrumentation for their interrogation. This situation could create a bottleneck in the development process without automation.
Upgrade to Plates
Break through your bottleneck by upgrading your DLS unit to the DynaPro™ DLS Plate Reader. The DynaPro DLS Plate Reader excels at high-throughput sizing and screening of pharmaceutical and nanoparticle formulations to enable novel workflows impossible with other DLS instruments.
Instead of measuring one cuvette at a time, the DynaPro Plate Reader measures DLS and SLS using industry-standard 96, 384, and 1536 well plates, allowing hundreds of sample screenings in a matter of hours. The flexible plate-based format easily integrates into any liquid handling system. An API software is available to interface the DynaPro Plate Reader for full automation integration. This capability allows for automated plate loading and data collection.
Even if you need to run just a handful of samples, upgrading to plates is a great productivity booster. Simply pipette samples into the plate, set up the measurement, and return when it's done. The hands-off workflow frees up your time to focus on data analysis and interpretation.
Flexible and Comprehensive
Complementing its high-throughput capabilities, the DynaPro Plate Reader stands out because of its low sample consumption and precise temperature control. With as little as a 4µL aliquot, you can get up to six stability metrics (Size, Polydispersity, Particle concentration, Turbidity, Tm, Tagg). The low volume requirement and comprehensive data enable you to save precious samples, particularly where sample availability is limited.
Early in the discovery process, the DynaPro Plate Reader can be deployed as a pre-screening tool before binding assays. Aggregates often bind non-specifically to a target and are detected as false positives. By identifying the presence of aggregates beforehand, DLS serves as a guardian against erroneous results.
Purification or buffer exchange steps can trigger self-association because of rapid pH or buffer change. Plate-based DLS proves instrumental in process development by quickly sampling these different conditions or monitoring changes over time.
The DynaPro Plate Reader provides invaluable insights on various stability metrics during formulation development. Its excellent thermal control includes information such as the onset of aggregation and accelerated storage assessment. Given its ability to measure hundreds of samples with minimal volume, users can screen various parameters from a single plate.
